Target Information
Thioredoxin reductases (TrxRs)는 셀레노시스테인(selenocysteine)을 포함하는 피리딘 뉴클레오타이드-디설파이드 산화환원효소 계열입니다.
모든 포유류 TrxR은 글루타티온 환원효소와 유사한 1차 구조를 가지며, 활성 부위(-Cys-Val-Asn-Val-Gly-Cys-)와 C-말단의 셀레노시스테인(SeCys)을 포함하는 확장 서열(-Gly-Cys-SeCys-Gly-)을 특징으로 합니다.
TrxR은 FAD 보조기, NADPH 결합 부위, 그리고 산화환원 활성 부위를 포함하는 호모다이머 단백질로, NADPH에서 전자를 FAD와 활성 부위의 디설파이드, 그리고 SeCys 중심으로 전달하여 thioredoxin 등의 기질을 환원시킵니다.
TrxR 패밀리는 55–58 kDa 크기의 세 가지 동형체(TrxR1, TrxR2, TrxR3)를 포함하며, 세포 내 산화 스트레스 방어 및 전사인자 조절 등 다양한 생물학적 과정에서 중요한 역할을 합니다.
