Product Specific Information
The V9 monoclonal antibody recognizes human Vimentin, a 57 kDa intermediate filament protein expressed in cells of mesenchymal origin, as well as in radial glia, immature glial cells, and pancreatic precursor cells. It is considered a marker of cardiac differentiation. In neural cells, vimentin expression is gradually replaced by neurofilaments. Surface expression has been observed on activated macrophages, platelets, apoptotic T cells, and neutrophils.
This antibody also recognizes vimentin from canine, rat, and chicken, but not from mouse.
Applications Reported: Western blotting, immunocytochemistry, and immunohistochemistry (IHC-F, IHC-P).
Applications Tested: Verified for IHC-P using high pH antigen retrieval at 1.33 µg/mL. Optimal dilution should be determined experimentally.
Usage Note: Before use, briefly centrifuge the conjugate solution and use only the supernatant to minimize nonspecific background caused by aggregates.
Target Information
Vimentin is a developmentally regulated intermediate filament protein (IFP) found in mesenchymal cells. It participates in intracellular protein transport between the nucleus and plasma membrane and is co-expressed with desmin during early development. Vimentin serves as a storage network for steroidogenic cholesterol-containing lipid droplets and is involved in steroid synthesis regulation. Phosphorylation of vimentin leads to filament breakdown and cytoskeletal rearrangements that facilitate cholesterol transport to mitochondria.
