Product Specific Information
PCH101 항체는 인간 FOXP3 단백질의 아미노 말단에 반응하며, scurfy(sf) 마우스에서 결함으로 확인된 forkhead/winged-helix 전사 조절 인자 가족의 구성원입니다.
CD4+CD25+ 조절 T세포(Treg)에서 FOXP3 mRNA가 높게 발현되며, CD4+CD25- 세포에서 FOXP3 발현 시 Treg 특성이 유도됩니다.
인간 PBMC를 FOXP3 염색 세트로 처리 시, 약 0.5~4% 림프구가 염색되며, 주로 CD25^bright 세포에서 관찰됩니다(기증자 변동 있음).
사용 전 단백질 결합체 용액을 마이크로센트리퓨지로 짧게 원심 분리 후 상등액만 사용하십시오. 이는 저장 중 형성된 단백질 응집체를 제거해 비특이적 배경 염색을 줄이는 데 도움이 됩니다.

FOXP3는 포크헤드/윙드헬릭스 전사 조절 인자 계열에 속하며, 면역 항상성 유지에 필수적인 단백질입니다.
CD25+CD4+ 조절 T세포에서 높은 수준으로 발현되고, CD4+CD25- 세포에서 낮은 수준, CD4- CD8+ 세포에서는 발현되지 않습니다.
FOXP3 유전자 결함은 마우스에서 scurfy 표현형을 유발하며, 인간에서는 IPEX 증후군(면역기능장애, 다내분비병증, 장병증, X-연관 증후군)과 관련 있습니다.
또한 FOXP3는 인간 유방암 및 대장암 세포에서 p53에 의해 DNA 손상 반응 시 조절됩니다.
