Product Specific Information
The monoclonal antibody 1A4 recognizes human, mouse, and rat alpha-smooth muscle actin (α-SMA), a 42 kDa protein that is a major component of the cytoskeletal structure. This antibody is specific for the alpha form of muscle actin, expressed by smooth muscle cells of blood vessels, myofibroblasts, and myoepithelial cells. It is also found in the parenchyma and stroma of various tissues.
Applications Reported:
- Immunohistochemical staining of formalin-fixed paraffin-embedded (FFPE) tissue sections
- Immunocytochemistry
Applications Tested:
- Tested by IHC of FFPE tissue using high pH antigen retrieval.
- Recommended concentration: 1 µg/mL (optimize for each assay).
Usage Note:
Before use, briefly centrifuge the conjugate solution and use only the supernatant to minimize nonspecific background from protein aggregates.
Target Information
Smooth Muscle Actin (SMA) belongs to the actin family, highly conserved proteins essential for cell motility, structure, and integrity.
- Alpha actin: major component of the contractile apparatus
- Beta and gamma actins: involved in regulation of cell motility
Smooth muscle actin is found in vessel walls, gut wall, myometrium, and myoepithelial cells. Mutations in the smooth muscle actin gene are associated with familial thoracic aortic aneurysm type 6.
During wound healing or fibrocontractive conditions, smooth muscle actin appears in fibroblastic stress fibers. Multiple alternatively spliced variants have been identified.
