Description:
The YKIX322.3 monoclonal antibody reacts with canine CD5 (Ly-1), a 67-kDa type I transmembrane glycoprotein expressed on thymocytes and mature T cells. NK cells in dogs also express CD5 at lower levels. In dogs, CD5 is not detectable on B cells or B cell lines.

Reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal canine peripheral blood cells. Recommended usage: 5 µL (0.125 µg) per test. A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL. Cell number may vary from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
APC-eFluor 780 emits at 780 nm and is excited with the Red laser (633–647 nm). Ensure your instrument can detect this fluorochrome.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vials and stained samples from light.

Fixation:
Samples can be stored in:

  (100 µL of cell sample + 100 µL of buffer)
• 1-step Fix/Lyse Solution (Product #00-5333-57)

Samples can be stored up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information

CD5 is a 67 kDa single-chain transmembrane glycoprotein expressed on human T-lymphocytes, most thymocytes, and many T-cell leukemias and lymphomas. It also reacts with a subpopulation of activated B-cells and may regulate T-cell proliferation. CD5 is expressed on 95% of thymocytes and 72% of peripheral blood lymphocytes, mainly in T cell areas of lymph nodes. Diseases associated with CD5 dysfunction include thymus cancer and Richter’s Syndrome.
