Target Information
The tachykinins are a family of small peptides that include the neurotransmitters substance P, neurokinin A, and neurokinin B, which act on three related but distinct seven-transmembrane G-protein coupled receptors.
The NK-3 receptor (NK3R) has greatest affinity for neurokinin B and is highly expressed in the supraoptic and paraventricular nuclei. Upon ligand binding, NK3R activates a phosphatidylinositol-calcium second messenger system, leading to vasopressin and oxytocin release.
NK3R may play roles in learning and memory, as knockout mice show cognitive deficits. It may also be involved in vagal afferent neuron regulation, though activation in airway nerves is likely mediated by substance P or neurokinin A.
