Target Information

TLR4 is a member of the Toll-like receptor (TLR) family, which plays a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved across species and share structural and functional similarities.
TLR4 is most abundantly expressed in the placenta and in the myelomonocytic subpopulation of leukocytes. Mammalian cells respond to LPS by activating TLR4, which is part of a multi-protein complex containing CD14, LY96, and TLR4.
This complex mediates signal transduction events induced by lipopolysaccharide (LPS) from gram-negative bacteria and aids in the recognition of pathogen-associated molecular patterns (PAMPs).
Mutations in the TLR4 gene have been associated with differences in LPS responsiveness. Several transcript variants of the TLR4 gene exist, though the protein-coding potential of most is uncertain.
TLR4 is expressed by peripheral blood monocytes, a small population of B-cells, and human placenta. Studies with TLR4-deficient mice indicate that the main ligand for TLR4 is LPS, and deficiency results in increased susceptibility to Gram-negative sepsis.
