Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor in the TNFR superfamily. It also exists as a soluble form (~26 kDa). Upon stimulation, FAS aggregates its intracellular FAS-associated death domains (FADD), forming the death-inducing signaling complex (DISC) and activating caspases. These caspases trigger a downstream cascade leading to apoptosis.
FAS is known to activate NF-kappaB, MAPK3/ERK1, and MAPK8/JNK pathways, and is involved in proliferative signaling in fibroblasts and T cells. Multiple alternatively spliced transcript variants encoding distinct isoforms have been identified; those lacking the transmembrane domain may negatively regulate apoptosis.
The Fas/Fas ligand system plays a role in diseases such as AIDS, hepatitis, and cancer, and is implicated in antiviral immune responses through induction of apoptosis.
