Product Specific Information
MA1-10029 detects Nsp1p from S. cerevisiae samples. To confirm high affinity for Nsp1p, strain RS453 (shortened isoform) was compared to wildtype strain BJ5465.
NSP1 in RS453 lacks six FXFG repeats, encoding a protein migrating at ~85 kDa on SDS-PAGE gels. Wildtype Nsp1p has a predicted MW of 86.5 kDa, typically migrating at ~100–108 kDa. SDS-PAGE results confirmed specificity for Nsp1p: wildtype at ~108 kDa and RS453 at ~91 kDa.
- Western Blot (WB): Use MA1-10029 diluted 1:10,000 (cell lysates) to 1:50,000 (nuclear fractions) with ECL detection.
- Other WB methods: Use 1:1,000–1:5,000.
- Immunofluorescence (IF): Use 1:1,000–1:10,000 on yeast cells.
Target Information
Nucleoporin NSP1 is a component of the yeast nuclear pore complex (NPC). NPC components (nucleoporins, NUPs) serve as structural elements and docking sites for nuclear transport factors.
Directional transport is facilitated by FG-repeat affinity gradients and GSP1/GSP2 GTPase gradients across the nuclear envelope.
NSP1 is essential for multiple nuclear transport pathways, including poly(A)+ RNA, tRNA, pre-ribosome, SRP, and protein transport.
