Target Information
TNF-alpha (Tumor Necrosis Factor alpha) is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF-alpha regulates immune cell activity, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It typically exists as multimers (dimers, trimers, pentamers) but shows a single 17 kDa band under non-reducing SDS-PAGE conditions.
Knockout studies in mice suggest TNF-alpha’s neuroprotective roles and tumor necrosis effects in tumor-bearing models. It also plays a role in immune responses to bacterial, viral, parasitic, and fungal infections, and contributes to tumor cell cytolysis in synergy with interferon-gamma. TNF-alpha has been implicated in autoimmune diseases, insulin resistance, and cancer.
