- Immunohistochemistry (PFA fixed):
Incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris Buffer with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
TNF alpha is a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers (two, three, or five noncovalently linked units) but shows a single 17 kDa band on SDS-PAGE under non-reducing conditions.
Knockout studies in mice suggest neuroprotective functions and tumor necrosis induction when injected into tumor-bearing mice.
TNF alpha also plays roles in immune responses to bacterial, viral, parasitic, and fungal infections and is implicated in autoimmune diseases, insulin resistance, and cancer.
