Target Information

ADRBK1 (GRK2) is a serine/threonine kinase involved in phosphorylation of G protein-coupled receptors (GPCRs). GRK2 (83 kDa) is one of six GRK family members and is ubiquitously expressed in mammals. Upon ligand binding, GPCRs are phosphorylated by GRKs, leading to internalization mediated by beta-arrestin-1 and activation of the Ras–Raf–ERK1/2 signaling pathway. GRK2 activity is regulated by phosphorylation via PKC, Src, and ERK1/2, as well as protein interactions. ERK phosphorylates GRK2 on serine 670, inactivating it through a negative feedback mechanism.
