Product Specific Information

The HB5 monoclonal antibody reacts with human CD21, an approximately 145 kDa type I transmembrane protein. CD21 is expressed by mature B cells, follicular dendritic cells, and a subset of epithelial cells. It serves as a receptor for complement component C3d and Epstein-Barr virus (EBV), and forms a complex with CD19 and CD81 involved in B cell activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Usage Notes:

• Use at 5 µL (0.05 µg) per test (100 µL final volume).
• Cell count range: 10⁵–10⁸ cells/test.
• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Recommended viability dyes: Invitrogen LIVE/DEAD Fixable Dead Cell stains.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

CD21 (complement receptor 2, CR2, C3D receptor, EBV receptor)
CD21 binds C3 complement fragments and plays a critical role in uptake and retention of immunocomplexes, survival of memory B cells, and maintenance of the humoral immune response. It acts as the Epstein-Barr virus receptor and is involved in prion targeting to follicular dendritic cells. Genetic variations in CD21 are associated with systemic lupus erythematosus type 9 (SLEB9). Alternative splicing generates multiple isoforms.
