Product Specific Information

The RPA-T8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as a CD8α/β heterodimer or CD8α/α homodimer. CD8a is found on most thymocytes and subsets of mature T and NK cells. It binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a key role in T-cell development and activation.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for human peripheral blood cells at 5 µL (0.02 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
    • Add 5 µL per sample before mixing with cells.
    • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family, essential in T-cell signal transduction, differentiation, and activation. It is expressed predominantly on T cells as a CD8α/β heterodimer, functioning as a co-receptor with the TCR for MHC class I molecules. CD8α is also found as a homodimer on subsets of T cells, NK cells, monocytes, and dendritic cells. Engagement with MHC-I/peptide complexes triggers Lck kinase recruitment, leading to cytokine production, motility, and activation of cytotoxic T lymphocytes (CTLs) for pathogen and tumor cell clearance. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
