Product Specific Information

The HB5 monoclonal antibody reacts with human CD21, an approximately 145 kDa type I transmembrane protein. CD21 is expressed by mature B cells, follicular dendritic cells, and a subset of epithelial cells. It functions as a receptor for complement component C3d and Epstein-Barr virus (EBV). CD21, along with CD19 and CD81, forms a multimolecular complex with BCR, contributing to B cell activation.

Each product contains:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry of normal human peripheral blood cells. Use 5 µL (0.05 µg) per test in 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance. Use 5 µL per sample for optimal results, including single-color controls.

NovaFluor conjugates utilize Phiton™ technology, featuring engineered nucleic acid dye structures for optimized fluorescent signatures and reduced spectral spillover.

Target Information

CD21 (complement receptor 2, CR2, C3D receptor, EBV receptor) binds C3 complement fragments and plays roles in:

• Uptake and retention of immune complexes
• Survival of memory B cells
• Maintenance of humoral response to T-dependent antigens
• EBV receptor function
• Prion targeting and neuroinvasion pathways

CD21 forms complexes with CD19, CD81, and Leu13, contributing to B cell signal transduction. Genetic variations in CD21 are associated with susceptibility to systemic lupus erythematosus type 9 (SLEB9). Alternative splicing produces multiple isoforms.
