Target Information
Signal-mediated nuclear import and export occur through the nuclear pore complex (NPC), composed of approximately 50 unique nucleoporins. The 98 kDa nucleoporin (NUP98) is derived from a 186 kDa precursor that is cleaved to yield both 98 kDa and 96 kDa nucleoporins localized to the nucleoplasmic side of the NPC. Studies indicate that NUP98 functions as a docking site nucleoporin for transport substrates.
The human NUP98 gene is implicated in several chromosomal translocations associated with acute myelogenous leukemia (AML) and T-cell acute lymphocytic leukemia (T-ALL). It resides in the imprinted gene domain of 11p15.5, a critical tumor-suppressor region linked to Beckwith-Wiedemann syndrome, Wilms tumor, rhabdomyosarcoma, adrenocortical carcinoma, and cancers of the lung, ovary, and breast. Alternative splicing produces multiple transcript variants, not all of which are fully characterized.
