Product Specific Information

The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, a subpopulation of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and is critical for T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck via its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Applications Tested: Tested on mouse splenocytes; ≤0.125 µg/test recommended.
A test is defined as the amount of antibody that stains a cell sample in 100 µL total volume. Optimal titration is advised.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the 488 nm blue laser. It can substitute for PerCP-Cyanine5.5. Recommended filter: 710/50 bandpass (695/40 acceptable).

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal signal loss. Clone-specific performance should be confirmed empirically.

Target Information

CD4 is a co-receptor for MHC class II molecules and HIV. It is primarily expressed on T helper lymphocytes and also found on monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. CD4 facilitates T-cell activation via association with the T-cell receptor complex and kinase Lck, and may mediate neuronal injury in immune-related CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
