Product Specific Information

The RM4-5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and is crucial for T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck via its cytoplasmic tail. Binding of RM4-5 is blocked by GK1.5.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse spleen cells; ≤0.125 µg/test recommended. A test is defined as antibody amount for staining a 100 µL cell sample. Cell count may vary (10⁵–10⁸ cells/test). Optimize dilution empirically.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure instrument compatibility.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) up to 3 days at 4°C in dark with minimal impact on brightness and FRET efficiency.

Target Information

CD4 antigen recognizes MHC class II molecules and serves as a co-receptor for HIV. It is mainly expressed in T helper cells but also found in monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut. CD4 participates in T-cell activation via association with the T-cell receptor complex and kinase Lck, and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
