Description:
The 8D4-8 antibody reacts with human interleukin-4 (IL-4), a 15–19 kDa cytokine secreted by Th2 cells.
Applications Reported:
This antibody is reported for use in intracellular staining followed by flow cytometric analysis.
Applications Tested:
Pre-diluted and validated for flow cytometric analysis of stimulated normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol.
Refer to Staining Intracellular Antigens for Flow Cytometry, Protocol A at www.thermofisher.com/flowprotocols.
Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test.
Fluorochrome Information:
eFluor 450 is an alternative to Pacific Blue, excited at 405 nm and emitting at 446 nm. Ensure the instrument is compatible with this fluorochrome.

Target Information
Interleukin-4 (IL-4) is a pleiotropic immune-modulatory cytokine produced by activated T cells and serves as a ligand for the IL-4 receptor. It has broad biological functions and shares overlapping roles with IL-13. STAT6 acts as a key mediator in IL-4 signaling.
The IL-4 gene, along with IL-3, IL-5, IL-13, and CSF2, forms a cytokine gene cluster on chromosome 5q. IL-4 and IL-13 are closely located and regulated by long-range elements over a 120 kb region. IL-4 delta 2 is a known splice variant.
