Target Information
STAT4 was originally identified using degenerate primers complementary to conserved regions of other STAT proteins. STAT4 shares high similarity with STAT1 (52%) and STAT3 (47%). It can be tyrosine phosphorylated by Jak1 or Jak2, forming homo- or heterodimers with other STAT members. Phosphorylated STAT4 binds the IFN-gamma activated site (GAS), and additional serine phosphorylation enhances transcriptional activity.
STAT4 expression is mainly found in the thymus, spleen, and testis. It is activated by interleukin-12 (IL-12), which is essential for IFN-gamma induction and Th1 cell differentiation. STAT4-deficient mice show impaired Th1 development and increased Th2 differentiation. STAT4 activation by IFN-α/β is also required for IFN-γ production during viral infection.
