Target Information
The munc-18 interacting protein (Mint) family consists of adaptor proteins involved in membrane transport and organization. In mammals, Mint1, Mint2, and Mint3 isoforms exist. The C-terminal half of these proteins contains conserved PTB and PDZ domains that mediate protein interactions.
Mint1 and Mint2 are brain-specific and bind to Munc18, while Mint3 is ubiquitously expressed and does not interact with Munc18. Mint3 interacts with Alzheimer’s Disease-related amyloid precursor protein (APP) through PTB and PDZ domains, potentially linking APP to transport machinery components and regulating its transport, endocytosis, and metabolism. Abnormal APP metabolism is associated with early-onset Alzheimer’s disease.
