Description:
The BVD6-24G2 antibody reacts with mouse interleukin-4 (IL-4), a 14 kDa cytokine secreted by Th2 cells.

Applications Reported:
This antibody is reported for use in intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and tested for intracellular staining and flow cytometric analysis of restimulated mouse splenocyte cultures. Recommended use: 5 µL (0.5 µg) per test.
A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically (10⁵–10⁸ cells/test).

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation; protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation.
Clone-specific performance should be determined empirically.

Excitation/Emission:
488–561 nm / 775 nm; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

Interleukin-4 (IL-4) is a pleiotropic, immune-modulatory cytokine produced by activated T cells and serves as a ligand for the interleukin-4 receptor.
IL-4 has a broad range of biological actions and shares overlapping functions with IL-13 through receptor binding. STAT6 mediates IL-4–induced immune regulatory signaling.
The IL-4 gene is located on chromosome 5q within a cytokine gene cluster (IL-3, IL-5, IL-13, CSF2) and is closely linked to IL-13.
IL-4, IL-5, and IL-13 are co-regulated by long-range regulatory elements spanning approximately 120 kb.
IL-4 delta 2 is a known splice variant.
