Target Information

BACE1 (beta-secretase) is a key enzyme involved in the production of amyloid beta-peptides (Abeta), which accumulate in extracellular amyloid plaques characteristic of Alzheimer’s disease (AD).
Mutations such as the “Swedish” mutation in amyloid precursor protein (APP) enhance cleavage by BACE1, contributing to early-onset familial AD.
Therapeutic research has focused on inhibiting BACE1 and other APP-cleaving enzymes like gamma-secretase.
APP is first cleaved by beta-secretase, generating a soluble derivative and a membrane-anchored 99-amino acid C-terminal fragment (C99).
C99 is then processed by gamma-secretase to produce the 4 kDa amyloid-beta peptide, which accumulates in AD brains.
BACE1 (also known as Asp2) is a transmembrane aspartic protease that colocalizes with APP and plays a central role in amyloidogenesis.
