Product Specific Information
The eBioH4A3 monoclonal antibody reacts with human CD107a, also known as lysosomal-associated membrane protein-1 (LAMP-1). CD107a is a highly glycosylated ~110 kDa protein expressed mainly in lysosomal/endosomal membranes of nearly all cells. It transiently appears on the cell surface of degranulating cytolytic T cells and is upregulated in activated platelets and certain cancer cells.

- Intracellular staining followed by flow cytometric analysis
- Surface staining in flow cytometric degranulation assays
Applications Tested:
Pre-titrated and tested by intracellular staining and flow cytometric analysis of Jurkat cells. Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test.

Target Information
LAMP1 (CD107a) is a major component of the lysosomal membrane, typically co-expressed with LAMP2. About 1–2% of CD107a is found on the plasma membrane. It is a heavily glycosylated type I integral membrane protein involved in lysosome biogenesis and fusion with phagosomes.
Upon activation, LAMP1 translocates to the plasma membrane via a carboxyl-terminal tyrosine-based motif (YXXI). Mutations affecting this motif alter lysosomal localization, causing cycling between plasma membrane and endosome. Cell surface LAMP1 and LAMP2 promote PBMC adhesion to vascular endothelium, suggesting roles in inflammation. Elevated LAMP1 expression is observed around senile plaques in Alzheimer’s disease and in various immune cells during thymic involution. LAMP1 serves as an effective marker of mast cell activation.
