Histone H2A.X (H2AX)는 핵심 히스톤 중 하나로, DNA 이중가닥 절단(DSB) 시 Ser139 위치의 인산화가 일어나 γ-H2AX를 형성합니다. 이 인산화는 Western blot 또는 면역형광으로 검출 가능하며, DSB 빈도를 반영합니다.
H2AX 인산화는 ATM, DNA-PK, ATR 등의 인산화 효소에 의해 조절됩니다.
Histone H2A.X는 143개의 아미노산으로 구성되며, G1 및 S-phase에서 합성되는 기본 히스톤입니다.
두 가지 형태의 mRNA(1600 bases, polyA 포함 / 575 bases, polyA 없음)가 존재하며, 각각 basal 및 replication type으로 작용합니다.
인간 염색체 11q23.2-q23.3 영역에 위치하며, DNA 구조 형성과 손상 복구 단백질 MDC1의 결합을 조절합니다.
