Target Information
SMAD2 (also known as MADH2 or MAD2) regulates multiple cellular processes such as cell proliferation, apoptosis, and differentiation. SMAD2 interacts with TGF-beta receptors through the SMAD anchor for receptor activation, and its translocation into the nucleus is central to TGF-beta signaling. Phosphorylation by ERK1 enhances SMAD2 transcriptional activity, while calmodulin negatively regulates it. Upon TGF-beta stimulation, SMAD2 is phosphorylated, dissociates from SARA, and associates with SMAD4 to form transcriptional complexes in the nucleus. SMAD2 can also mediate activin signaling via phosphorylation by activin type 1 receptor kinase. Multiple alternatively spliced transcript variants have been identified.
