Target Information
Smad4는 인간 염색체 18번에 위치한 Smad4 유전자에 의해 암호화됩니다. Smad4는 Smad family member 4, Mothers against decapentaplegic homolog 4 등으로도 알려져 있습니다.
Smad4 단백질은 N-말단의 MH1 (MAD homology 1) 도메인과 C-말단의 MH2 (MAD homology 2) 도메인을 가지며, 각각 DNA 결합과 단백질 상호작용에 관여합니다.
TGF-beta 및 BMP 리간드가 수용체에 결합하면 R-Smad(1,2,3,5,8)가 인산화되고, Smad4(co-Smad)와 삼합체를 형성하여 핵으로 이동합니다. Smad6, Smad7 등의 I-Smad는 이러한 신호전달을 억제합니다.
Smad4는 Cyclin D1 억제 및 collagen 발현 촉진을 통해 세포 증식을 억제하는 종양 억제 단백질로 작용합니다. 탈인산화 과정은 Smad의 핵 내외 이동을 조절합니다.
