Description:
The 4H11 monoclonal antibody reacts with human CD34, also known as mucosialin. CD34 belongs to a protein family that includes endoglycan and podocalyxin. These are single-pass transmembrane proteins with heavily glycosylated extracellular mucin domains. CD34 was first identified as an antigen on hematopoietic progenitors and is widely used as a marker to isolate cells capable of hematopoietic engraftment.
The 4H11 antibody belongs to class III epitopes, which react with a protein epitope.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use is 5 µL (0.5 µg) per 100 µL test volume (10⁵–10⁸ cells/test).
Excitation: 488–561 nm

Target Information
CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on many stem cell populations. It serves as a marker for stem cells and may function as a receptor regulating adhesion, differentiation, and proliferation. CD34 expression reflects specific hematopoietic developmental stages and may mediate attachment to the bone marrow matrix or stromal cells.
It may also act as a scaffold for lineage-specific glycans, facilitating binding to lectins expressed by stromal or marrow components. The intracellular domain is phosphorylated by protein kinase C, suggesting a role in signal transduction.
