Product Specific Information

The C17.8 antibody reacts with the p40 subunit of IL-12 and IL-23. It neutralizes IL-12 and IL-23 bioactivity. For specific neutralization of IL-23 only, clone G23-8 (anti-mouse IL-23 p19) is recommended.

IL-12 is a 70 kDa heterodimeric cytokine composed of two chains, p40 and p35, secreted by activated monocytes, macrophages, and dendritic cells. IL-23 is a heterodimer composed of p19 and p40 proteins, expressed by activated dendritic cells.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested by intracellular staining and flow cytometric analysis of stimulated mouse cells at ≤0.125 µg/test.
A test is defined as the amount of antibody that stains a sample in 100 µL volume. Cell number may range from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

eFluor® 660 is a replacement for Alexa Fluor® 647. It emits at 659 nm and is excited with a 633 nm red laser. Ensure your instrument can detect this fluorochrome.

Target Information

IL-12 is an interleukin produced by dendritic cells, neutrophils, macrophages, and lymphoblastoid cells upon antigenic stimulation. It is a heterodimeric cytokine encoded by IL-12A (p35) and IL-12B (p40).
IL-23 shares the p40 subunit with IL-12 and includes an IL-23A (p19) subunit.

Functions:

• IL-12 increases endothelial adhesion molecule-1 expression and directs leukocytes toward tumors.
• IL-23 stimulates memory T-cells and supports Th17 cell function, promoting neutrophil chemokine and pro-inflammatory cytokine expression.
