Product Specific Information

The monoclonal antibody MH9A4 recognizes interleukin-9 (IL-9), a proinflammatory cytokine involved in type 2 immune responses. Recent studies indicate IL-9 may be secreted by a distinct T-helper lineage called Th9, derived from Th2 cells with TGF beta or from naive CD4+ T cells with TGF beta and IL-4.

IL-9 belongs to the common cytokine receptor gamma chain-dependent family, which includes IL-2, IL-4, IL-7, IL-15, and IL-21. It affects Th2 lymphocytes, B lymphocytes, mast cells, eosinophils, and epithelial cells, linking IL-9 to asthma and allergy-related disorders.

Staining with MH9A4 antibody is not blocked by MH9D1, indicating distinct epitopes.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining and flow cytometric analysis of stimulated human peripheral blood cells. Use at 5 µL (0.06 µg) per test in a final volume of 100 µL; cell number range 10⁵–10⁸ cells/test.

Target Information

Interleukin 9 (IL-9) is produced by activated T helper 2 (Th2) lymphocytes. It regulates various cell types involved in asthma responses, including B and T lymphocytes, mast cells, eosinophils, and epithelial cells. Mouse IL-9 cDNA encodes a 144-amino-acid precursor with an 18-amino-acid signal peptide, yielding a mature cysteine-rich protein (~14 kDa). Native mouse IL-9 is heavily glycosylated. TGF beta promotes differentiation of Th17 and regulatory T cells and reprograms Th2 cells to secrete IL-9, driving Th9 cell formation.
