The monoclonal antibody 44.189 reacts with human CD336 (NKp44/NCR2), a 44-kDa transmembrane glycoprotein of the natural cytotoxicity receptor (NCR) family containing an ITIM domain.
CD336 expression and NK cell activation by this receptor require association with DAP12, an ITAM-containing transmembrane accessory protein.

Originally thought to be expressed only on IL-2-activated NK cells and γδ T lymphocytes, CD336 has since been found on IL-3-stimulated plasmacytoid dendritic cells, αβ T cells in the small intestinal epithelium, and T cells in cord blood.
CD336 is involved in the innate response of activated NK cells against viral infection and tumor cell proliferation.

The 44.189 antibody facilitates CD336 crosslinking and is reported for use in flow cytometric analysis.
It has been pre-titrated and tested by flow cytometric analysis of stimulated normal human peripheral blood cells.
Use at 5 µL (0.125 µg) per test, where one test corresponds to staining a sample in a final volume of 100 µL.
Cell number can range from 10⁵ to 10⁸ cells/test.
