Cofactors A와 D는 β-tubulin 중간체를 안정화시키는 역할을 하며, Cofactor E는 cofactor D/β-tubulin 복합체에 결합합니다. 이후 cofactor C의 작용으로 β-tubulin 폴리펩타이드가 방출되어 native 상태로 전환됩니다.
