Product Specific Information
The 11B11 antibody reacts with mouse interleukin-4 (IL-4), a 14 kD cytokine, and functions as a neutralizing antibody. IL-4, also known as BCDF, BCGF-1, and BSF-1, is secreted mainly by activated Th2 and natural killer T cells, and promotes B cell proliferation and differentiation. It also induces Ig isotype switching and upregulates MHC class II and CD23 expression.
Applications Reported:
Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested:
Validated for intracellular staining of mouse Th2-polarized splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
Recommended usage ≤ 0.25 µg per test (100 µL final volume). Cell numbers from 10⁵–10⁸ cells/test are typical. Optimize antibody concentration for best performance.
Fluorochrome Information:
Brilliant Violet™ 480 emits at 479 nm and is designed for cytometers equipped with a 405 nm violet laser. Ensure compatibility before use.
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to reduce non-specific interactions.
Not compatible with methanol-based fixation.

Target Information
Interleukin-4 (IL-4) is an immune-modulatory cytokine produced by activated T cells, acting as a ligand for the IL-4 receptor. It shares functional overlap with IL-13 due to receptor binding. STAT6 mediates IL-4 signaling. The IL-4 gene is located on chromosome 5q near IL-13 and IL-5, forming a cytokine cluster regulated over ~120 kb. IL-4 delta 2 is a known splice variant.
