Target Information
Inactive complement component 3b (iC3b) is a 42 kDa neoantigen present in blood serum. Cleavage of C3b into iC3b by the protease factor I, in the presence of cofactors, alters its structure and binding properties, preventing interaction with complement factor B and properdin. This regulation halts further progression of the complement cascade.
iC3b is thought to play a role in immune tolerance by binding complement receptor type 3 on antigen-presenting cells, stimulating production of transforming growth factor beta2 and interleukin-10. It also interacts with B-cells via the CD21 receptor, linking innate and adaptive immunity. Additionally, iC3b acts as an opsonin recognized by complement receptor 3 on leukocytes or CRIg on Kupffer cells, promoting phagocytosis of tagged cells.
Elevated iC3b levels are associated with autoimmune diseases such as Systemic Lupus Erythematosus and Rheumatoid Arthritis.
