Product Specific Information
The monoclonal antibody MH99 recognizes human CD326, also known as EpCAM (Epithelial cell adhesion molecule). This 40 kDa type I transmembrane protein is involved in calcium-independent cell-cell interactions and is expressed primarily on the basolateral surface of most epithelia.
Normal epithelia express low levels of CD326, but increased expression correlates with proliferation and progression to a mesenchymal phenotype. CD326 serves as a diagnostic marker for circulating metastatic carcinoma cells, while non-epithelial cancer cells do not express EpCAM.
Based on crossblocking studies, MH99 and 1B7 recognize overlapping epitopes.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of A549 cell line. Recommended usage is 5 µL (0.5 µg) per test (100 µL final volume).
Excitation: 488 nm; Emission: 519 nm; Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Ep-CAM (epithelial adhesion molecule, epithelial specific antigen, ESA) is a transmembrane glycoprotein (~40 kDa) expressed in epithelium. It functions as a homotypic calcium-independent cell adhesion molecule and influences cell cycle, proliferation, and metabolism by inducing c-myc, cyclin A, and E.
Ep-CAM-mediated adhesions negatively regulate cadherin-mediated adhesions, affecting epithelial differentiation and growth. Overexpression is associated with enhanced epithelial proliferation and is highly expressed in human carcinomas, serving as a marker for epithelial lineage tumors.
Ep-CAM is found on basolateral surfaces of most simple epithelia and many carcinoma types, and can distinguish adenocarcinomas from pleural mesotheliomas.
