Product Specific Information

This 8CH monoclonal antibody reacts with human IL-8 (CXCL8), a pro-inflammatory CXC chemokine. IL-8 is synthesized as a 99 amino acid precursor, processed into isoforms of 72 or 77 amino acids. IL-8(77), secreted mainly by endothelial cells, is less potent as a neutrophil activator and plays a role in angiogenesis during fetal development. IL-8(72), predominant in adults, is expressed by immune and epithelial cells under inflammatory conditions and mediates neutrophil activation and migration.
Signaling occurs via CXCR1 and CXCR2 receptors. IL-8 expression is often elevated in tumors, contributing to angiogenesis and metastasis.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and tested using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
Use at 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Excitation: 488 nm; Emission: 519 nm; Laser: Blue.

Target Information

Interleukin 8 (IL-8, CXCL8) is a 72 amino acid pro-inflammatory factor in the CXC chemokine family, binding to IL-8RA and IL-8RB receptors. It functions as a chemoattractant and angiogenic factor, induced by inflammatory stimuli in macrophages and endothelial cells. In endothelial cells, IL-8 is stored in Weibel-Palade bodies.
It signals through CXCR1 and CXCR2, and is also known as NAP-1, GCP-1, MONAP, and 3-10C. IL-8 and related chemokines form a gene cluster on chromosome 4q. IL-8 plays roles in tumor angiogenesis, growth, and bronchiolitis pathogenesis.
