Product Specific Information

This FL34Z3L monoclonal antibody reacts with human CCL4, also known as MIP-1β (Macrophage Inflammatory Protein 1 beta).
CCL4 is a member of the CC-chemokine family and is closely related to CCL3 (MIP-1α). These proteins play critical roles in leukocyte recruitment to sites of inflammation.
CCL4 preferentially attracts CD4+ T cells, while CCL3 mainly attracts CD8+ T cells. In addition, CCL4 induces proinflammatory cytokine release, mast cell degranulation, and NK cell activation.

CCL4 signaling occurs via G protein-coupled receptors CCR1, CCR4, and CCR5. CCR5 serves as a co-receptor for HIV entry, and its ligands, including CCL4, inhibit viral entry by blocking gp120-CCR5 interaction and promoting receptor internalization.

  • Surface and intracellular staining for flow cytometric analysis.

Applications Tested:

  • Pre-diluted and tested for intracellular staining of stimulated human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
  • Use at 5 µL (0.5 µg) per test (100 µL final volume).
  • Suitable for 10⁵–10⁸ cells/test.

Target Information

CCL4 (macrophage inflammatory protein 1-beta, MIP1B) belongs to the CC chemokine family.
It is produced by macrophages upon bacterial endotoxin stimulation and recruits various inflammatory cells to sites of inflammation.
CCL4 and CCL3 regulate immune cell trafficking and activation during infection by bacteria, viruses, parasites, and fungi.
CCL4 selectively attracts CD4+ lymphocytes, while CCL3 attracts CD8+ lymphocytes, and both act as chemoattractants for B cells, eosinophils, and dendritic cells.
Processed CCL4 can down-regulate CCR5 expression, inhibiting HIV-1 entry into T cells.
CCL4 binds with high affinity to CCR5 receptors.
