Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

MMP9 (matrix metallopeptidase 9, GELB, CLG4B)는 아연 및 칼슘 의존성 엔도펩티다아제 계열에 속하는 단백질로, 세포외기질 단백질을 분해하는 역할을 합니다.
MMP9는 92 kDa 전구체(zymogen) 형태로 분비되며, 절단되어 활성형 효소(82 kDa)를 형성합니다.
이 단백질은 다음과 같은 도메인을 포함합니다:

• 젤라틴 결합 도메인 (fibronectin type II units 3개)
• 아연 결합 부위를 포함한 촉매 도메인
• 프롤린 풍부형 type V collagen-homologous 도메인
• hemopexin-like 도메인

MMP9는 단핵구, 대식세포, 호중구, 각질형성세포, 섬유아세포, 파골세포 및 내피세포 등 다양한 세포에서 발현되며, 염증 반응, 조직 재형성, 상처 치유, 종양 성장 및 전이에 관여합니다.
또한 MMP9는 type IV 및 V 콜라겐을 분해하며, IL-8에 의해 유도되는 조혈모세포의 골수 이동 및 종양 관련 조직 재형성에도 역할을 하는 것으로 알려져 있습니다.
높은 MMP9 발현은 특발성 심방세동과 대동맥류의 진행과 관련이 있습니다.
