• For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

MMP9 (matrix metallopeptidase 9, GELB, CLG4B) is a zinc- and calcium-dependent endopeptidase that degrades extracellular matrix proteins.
It is secreted as a 92 kDa zymogen and becomes active (82 kDa) after cleavage.
MMP9 contains:

• A gelatin-binding domain with three fibronectin type II units
• A catalytic domain with a zinc-binding site
• A proline-rich type V collagen-homologous domain
• A hemopexin-like domain

MMP9 is produced by monocytes, macrophages, neutrophils, keratinocytes, fibroblasts, osteoclasts, and endothelial cells.
It plays key roles in inflammatory responses, tissue remodeling, wound healing, tumor growth, and metastasis.
Elevated MMP9 levels have been linked to idiopathic atrial fibrillation and aortic aneurysm progression.
