Product Specific Information

• Tested in direct ELISA (for coating use antibody at 1–5 µg/mL).
• For immunohistochemistry (PFA-fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

ATG5 (Autophagy Related 5) is a critical component of the autophagy process and plays an important role in apoptosis. It is involved in mitochondrial quality control, negative regulation of the innate antiviral immune response, lymphocyte development, MHC II antigen presentation, and adipocyte differentiation.
Following conjugation with ATG12, the ATG5–ATG12 complex participates in autophagosome formation. ATG5 contributes to autophagic cell death by interacting with Fas-associated protein with death domain (FADD).
The ATG5–ATG12 conjugate forms a cup-shaped isolation membrane that detaches before or after autophagosome completion.
ATG5 also associates with innate immune response proteins such as RIG-I and VISA (IPS-1), inhibiting type I interferon production and facilitating viral replication.
Dysfunction of ATG5 is associated with diseases such as spinocerebellar ataxia.
