Product Specific Information
This antibody binds to carboxy-terminal oligo-histidine tags (6x His tags) of fusion proteins. It also has affinity for internal His-tags but no affinity for N-terminal His-tags.
Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH). It is commonly expressed at either N- or C-terminal regions of recombinant proteins to facilitate purification by immobilized metal affinity chromatography (IMAC).
Epitope tagging enables detection of recombinant proteins by fusing a known epitope to the protein of interest. His-tagged proteins can be purified using metal chelate chromatography due to the strong and selective binding of histidine residues to metal ions such as Ni²⁺. His tag-specific antibodies are used to detect, isolate, or co-immunoprecipitate tagged proteins.
