This antibody binds to carboxy-terminal oligo-histidine tags (6x His tags) of fusion proteins. It also has affinity for internal His-tags but no affinity for N-terminal His-tags.

Target Information

The 6x His tag is a synthetic oligo peptide consisting of six consecutive histidine residues (HHHHHH).
It is commonly expressed at either N- or C-terminal regions of recombinant proteins to enable isolation or purification via immobilized metal affinity chromatography (IMAC).

Epitope tagging allows detection of recombinant proteins by fusing known epitopes, making it possible to identify proteins lacking specific antibodies. His-tagged proteins are purified using metal chelate chromatography due to the strong and selective affinity of histidine clusters for Ni²⁺ ions, typically on Ni²⁺-NTA adsorbents.

His tag-specific antibodies are used for detection or co-immunoprecipitation of tagged proteins.
