Product Specific Information

The 4S.B3 monoclonal antibody reacts with interferon-gamma (IFN gamma). Human IFN gamma is a 17 kDa cytokine produced by activated T and NK cells, exhibiting antiviral and antiparasitic activity. It acts synergistically with cytokines such as TNF alpha to inhibit cell proliferation and modulate immune responses across various cell types expressing IFN gamma receptors. Glycosylation does not affect its biological activity.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and validated for intracellular staining of stimulated human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) and standard flow cytometry protocols.
Recommended use: 5 µL (0.015 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorophore Information:
Alexa Fluor 660 emits at 690 nm, suitable for spectral cytometers and multiplexing with APC, Alexa Fluor 647, and Alexa Fluor 700.

Target Information

Interferon gamma (IFN-γ, Type II interferon) is a macrophage activation factor produced mainly by T-lymphocytes and NK cells in response to antigens and cytokines. It functions as a homodimer (~45 kDa) and exhibits antiviral, antiproliferative, and immunomodulatory activities. IFN-γ induces MHC class I and II expression, activates macrophages, and enhances immunoglobulin secretion by B cells. Activity is species-specific; human IFN-γ does not cross-react with mouse IFN-γ. Activation occurs via IFN-γ receptor I/II and the JAK-STAT pathway. Expression is upregulated by IL2, FGF basic, and EGF, and downregulated by vitamin D3 or DMN. Mutations in the IFN-γ gene are associated with aplastic anemia.
