Background

This gene encodes the alpha chain of type XVIII collagen, one of the multiplexins—extracellular matrix proteins containing multiple triple-helix domains interrupted by non-collagenous regions. The long isoform includes an N-terminal domain homologous to frizzled receptors. Proteolytic processing at several cleavage sites in the C-terminal region produces endostatin, a potent antiangiogenic protein that inhibits angiogenesis and tumor growth. Mutations in this gene are associated with Knobloch syndrome, characterized by retinal abnormalities, suggesting type XVIII collagen’s role in retinal structure and neural tube closure.
