Background

  • Domain: Intact BH3 motif is required for pro-apoptotic activity and interaction with Bcl-2 family proteins (BIK, BID, BAK, BAD, BAX).
  • Function: The major proteolytic product p15 BID promotes cytochrome c release. Isoforms 1, 2, and 4 induce apoptosis, while isoform 3 does not.
  • Post-translational modification (PTM): Phosphorylated upon DNA damage (likely by ATM or ATR). TNF-α induces caspase-mediated cleavage of p22 BID into p15, p13, and p11 products.
  • Subunit: Forms heterodimers with pro-apoptotic BAX or anti-apoptotic Bcl-2.
  • Tissue Specificity:
    • Isoforms 2 and 3: spleen, bone marrow, cerebral and cerebellar cortex
    • Isoform 2: spleen, pancreas, placenta
    • Isoform 3: lung, pancreas, spleen
