FOXP3 (Forkhead box protein 3)는 포크헤드/윙드헬릭스 전사조절인자 계열에 속하며, 포유류에서 고도로 보존되어 있고 정상 면역 항상성 유지에 필수적인 단백질입니다.
FOXP3는 381개의 아미노산으로 구성되어 있으며, CD25⁺ CD4⁺ 조절 T세포에서 안정적으로 높은 수준으로 발현됩니다.
CD4⁺/CD25⁻ 세포에서는 낮은 수준으로, CD4⁻/CD8⁺ T세포에서는 발현되지 않습니다.
FOXP3는 조절 T세포의 주요 마스터 조절 유전자로 알려져 있으며, FOXP3 유전자 결함은 마우스에서 scurfy 표현형을 유발합니다.
인간에서는 FOXP3 결함이 IPEX 증후군(면역 이상, 다내분비병증, 장병증, X-연관 증후군)과 관련이 있습니다.
또한 인간 유방암 및 대장암 세포에서 FOXP3의 발현은 DNA 손상에 반응하여 p53에 의해 조절됩니다.
