Target Information

Modification of target proteins by ubiquitin participates in diverse biological functions. Proteins destined for degradation via the 26S proteasome are coupled to multiple ubiquitin copies, while ubiquitin or related molecules can also alter subcellular localization or protein activity. Deubiquitination, catalyzed by deubiquitinating enzymes (DUBs), includes four families: ubiquitin C-terminal hydrolases, ubiquitin-specific processing proteases (USPs), OTU-domain ubiquitin-aldehyde-binding proteins, and Jab1/Pad1/MPN-domain-containing metallo-enzymes. Among these, USPs are the most prevalent and release ubiquitin from conjugated proteins, featuring conserved catalytic domains with Cys and His boxes and variable extensions for substrate recognition and protein interactions.
