Preparation:
Produced from sera of goats immunized with highly pure Recombinant Human TNF-alpha. Anti-Human TNF-alpha-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect Human TNF-alpha by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, used with PeproTech Polyclonal Anti-Human TNF-alpha (500-P31AG) as a capture antibody, enables detection of at least 0.2–0.4 ng/well of Recombinant Human TNF-alpha.

Western Blot:
To detect Human TNF-alpha by Western Blot analysis, use 0.1–0.2 µg/mL of this antibody. When combined with compatible secondary reagents, the detection limit for Recombinant Human TNF-alpha is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune responses, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers (dimers, trimers, or pentamers) but shows a single 17 kDa band in SDS-PAGE under non-reducing conditions. Knockout studies in mice suggest its neuroprotective function and tumor necrotic activity. TNF alpha is also involved in immune responses against bacterial, viral, parasitic, and fungal infections, as well as tumor necrosis.
This cytokine has been implicated in autoimmune diseases, insulin resistance, and cancer.
