Description:
The dG9 antibody clone reacts with human perforin (pore-forming protein, pfp). Perforin is a cytolytic mediator present in cytoplasmic granules of cytotoxic T lymphocytes (CTL) and natural killer (NK) cells. It plays a key role in immune responses against tumors and viral infections.

Applications Reported:
This antibody is suitable for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for intracellular staining using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) following BestProtocols® Protocol A.
Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emitting at 445 nm. Ensure the instrument is compatible with Violet laser detection.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

Perforin is a major cytolytic protein stored and released by cytotoxic lymphocytes. It forms transmembrane pores similar to complement component C9 and is crucial for T-cell and NK-cell mediated cytolysis.
Defects in the perforin gene cause familial hemophagocytic lymphohistiocytosis type 2 (HPLH2), a rare autosomal recessive disorder.
Alternative splicing results in multiple transcript variants.
