Product Specific Information

The MIH1 monoclonal antibody reacts with human B7-H1 (PD-L1), a member of the B7 family and Ig superfamily with an approximate molecular weight of 40 kDa. PD-L1 is expressed on a majority of leukocytes and serves as a ligand for PD-1, leading to inhibition of T and B cell responses. MIH1 is reported to be a blocking antibody.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-titrated and validated for use at 0.8 µg per test
• Definition of Test: Amount of antibody sufficient to stain a sample in 100 µL final volume (10⁵–10⁸ cells/test)

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample prior to combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

Programmed death receptor ligand 1 (PD-L1, B7-H1) is a B7 family member that binds to PD-1, negatively regulating T-cell receptor (TCR) signaling. PD-L1 engagement reduces TCR-mediated proliferation and cytokine production. It is abundantly expressed in many human and murine cancers and upregulated by IFN-γ, playing a role in tumor immune evasion.
